Integrating Microbubbles into Hybridoma Workflows

Monoclonal antibodies (mAbs) are critical biomolecules with applications that span research, diagnostics, therapeutics, and beyond. Hybridoma technology, first discovered in 1975, has quickly become the preferable method of mAb production1. Generally speaking, the hybridoma workflow is laborious and resource-intensive, and has not seen many improvements or optimizations since its inception.

Hybridoma technology involves the fusion of cells from immunized hosts with immortalized myeloma cells to create a hybridoma. These hybridomas are then screened to identify the high-producing clones that yield antibodies with superior antigen specificity, and these selected clones are then expanded to increase production.

Akadeum’s Mouse B Cell Isolation Kit can be used to optimize the purification stage, enriching splenocyte samples for highly viable B cells for use in generating hybridomas.

Akadeum’s Mouse B Cell Isolation Kit delivers a high-purity population of healthy, viable mouse B cells using a protocol that is fast, easy, and exceptionally gentle on cells. Ideal for the enrichment of delicate immune cells, separation occurs directly in the sample container without exposing cells to harsh chemicals or harmful shear forces.

How it Works: First, Akadeum’s biotinylated antibody cocktail is added to the sample to label the contaminating cells. Next, the microbubbles are mixed into the sample where the seek out and bind to the unwanted cells. Once bound, they quickly and gently float the undesired cells to the surface. An optional centrifugation step pellets the B cells, and the microbubbles, microbubble-bound cells, and supernatant are easily aspirated away leaving the untouched and enriched B cells happy, healthy, and ready for downstream use.
