Human T Cell Leukopak Isolation Kit – GMP Grade

With quality standards built into the design and manufacturing process at every step, use our Human T Cell Leukopak Isolation Kit – GMP Grade to isolate truly untouched T cells directly from leukapheresis material without the need for lysis or density gradient centrifugation. The Human T Cell Leukopak Isolation Kit – GMP Grade was developed with BACS™ Microbubbles to negatively select untouched T cells from leukopak material. Non-T cells are targeted and removed with antibodies recognizing CD14, CD16, CD19, CD20, CD36, CD56, CD123, CD235ab via negative selection. Isolated T cells are suitable for a wide variety of downstream assays, including cell culture, flow cytometry, scRNA-Seq, or other functional studies. The Human T Cell Leukopak Isolation Kit – GMP Grade has the capacity to isolate T cells from an average half-leukopak (12.5 x 10⁹ total cells).

  • GMP grade – Our GMP grade allows you to have peace of mind, with stringent documentation and processes, traceability, and strict adherence to standards.
  • Enjoy higher recovery – Industry-leading recovery of T cells from leukopak material allows you to get the most out of your leukopaks, limiting donor recalls.
  • Shorter processing time – From leukopak bag to purified cells in under an hour enables greater productivity, adding flexibility to your day.
  • No dedicated equipment, no magnets, or columns – Only general tools and equipment are needed for our quick and easy workflow.

Product Details

The Human T Cell Leukopak Isolation Kit – GMP Grade streamlines T cell isolations by eliminating the need for excess equipment and time-consuming sample preparation (i.e., Ficoll gradients or lysis)—allowing you more time to spend focusing on what matters, your science. By eliminating the need for pre-processing, the Human T Cell Leukopak Isolation Kit – GMP Grade minimizes handling steps and consistently delivers more isolated T cells. Get the most out of your time and material with the Human T Cell Leukopak Isolation Kit – GMP Grade.

Mechanism of Action: Leukopak samples contain a milieu of immune cells (i.e., white blood cells), plasma, platelets, and red blood cells. The Direct from Human T Cell Leukopak Isolation Kit – GMP Grade uses a negative selection method to enrich T cells by removing unwanted cell populations. These unwanted cells are labeled with biotin-CD14, CD16, CD19, CD20, CD36, CD56, CD123, CD235ab. BACS™ Streptavidin Microbubbles are mixed into the sample where they bind to the labeled cells and gently float them to the surface. Untouched T cells in their original state are subsequently harvested.

How it Works: The Human T Cell from Leukopak Biotin-Antibody Cocktail is added to the leukapheresis product to label unwanted cell populations. BACS™ Streptavidin Microbubbles are added to the sample and bind to the biotin-labeled cells. Due to their inherent buoyancy, the microbubbles quickly and gently float the non-T cells to the surface. Following centrifugation, the T cells are collected.

Key Features

The Human T Cell Leukopak Isolation Kit – GMP Grade is designed to enrich T Cells from leukopaks. Akadeum’s novel Buoyancy Activated Cell Sorting (BACS™) Microbubbles enable processing, limiting the unnecessary loss of cells. The T cell isolation protocol is fast and gentle on cells, taking just one hour and not requiring upfront processing or magnets or columns. The result is a population of highly pure T cells that are ready for downstream use.

Elevated Solutions for Sample Preparation

  • Ficoll- and lysis-free cell separation for the isolation of truly untouched T cells.
  • Fast and easy workflow; leukopak bag to cells in under 60 min.
  • Maximum yield of T cells without compromising on high purity.

Elegant in Simplicity, Powerful in Application

BACS™ Microbubble technology harnesses the power of flotation to provide a fast, easy, and gentle T cell isolation without the need for pre-processing of the leukopak material. Our novel technology couples performance, ease, and quality, bringing cell separation to the next level.

Experience the Benefits of a Microbubble Workflow. Akadeum’s Microbubbles Are:

  • Gentle on cells: Harnessing the power of buoyancy, BACSTM Microbubbles swiftly and gently remove unwanted cells via flotation.
  • Fast, reliable, and scalable: The simple BACSTM Microbubbles protocol delivers reproducible results that can be easily scaled for greter numbers of samples or larger volumes.
  • Equipment-free: Free yourself from worrying about maintaining magnets, losing cells to clogged columns, or sacrificing space in the biosafety hood.

Untouched T cells were isolated from leukopak material using the Human T Cell Leukopak Isolation Kit – GMP Grade. Isolated T cells were labeled with CD45-FITC and CD3-APC Fire/750. The fluorescently labeled cells were analyzed by flow cytometry.  Dead cells and debris were excluded from the analysis.
