Akadeum Life Sciences has partnered with a cell therapy AlerionTM Microbubble Cell Separation System beta customer to help their team develop an improved method for T cell banking. The company faces significant challenges in recovery, purity, and processing time with its existing manual methods, and Akadeum’s AlerionTM is well suited to overcome the challenges with its proven superior cell recovery, purity, and scalability.

The Challenge

To develop and deliver effective CAR T therapies, the company needed to bank a large number of purified T cells efficiently. Their traditional manual processing method was time-consuming, resulting in suboptimal T cell recovery.

Finding a Solution With Akadeum’s AlerionTM System

To address these challenges, the company became a beta user of Akadeum’s AlerionTM Microbubble Cell Separation System. The system, specifically paired with Akadeum’s Human T Cell Leukopak Isolation Kit and the BioRiseT

M consumable, promised a more efficient process for delivering more T cells faster.

The research team processed three leukopak samples to demonstrate the AlerionTM’s real-world potential. Flow cytometry measured the percent concentration of TCR+ cells pre- and post-processing, and the results were used to calculate T cell recovery rates.

Discussion

The AlerionTM Microbubble Cell Separation System  demonstrated substantial improvements for T Cell Isolation:

  • Superior purity: As demonstrated in the flow cytometry data, the percentage of TCR+ cells post-processing improved for the beta user. The AlerionTM Microbubble Cell Separation System consistently enables higher purity T cells for better downstream cell therapy engineering.
  • Increased T cell recovery: The AlerionTM Microbubble Cell Separation System significantly boosted the percentage of T cell recovery. Leukopak samples 1 and 2 processed with the AlerionTM system achieved T cell recovery rates of 71.3% and 72.4%, respectively. This enables the banking of more T cells for downstream cell therapy applications from a single run.

With Akadeum’s AlerionTM Microbubble Cell Separation System, the company successfully enhanced its T cell isolation process. The improved recovery rates and reduced processing times allowed them to bank more T cells for their pilot cell therapy trials.
